The Separation Of Hydrolysate Of Enzymesoluble Pigskin By Chromatography And Sodium Dodecyl Sulphate-Polyacrylamide Gel Electrophoresis

Abstract

We report here on low pressure ion-exchange chromatography (LPIEC) with phosphate buffer solution eluent and Sodium Dodecyl Sulphate-Polyacrylamide Gel Electrophoresis (SDS-page) for separation of the hydrolysate of enzyme-soluble pigskin collagen. The experimental methods for LPIEC are described in detail. Using this method, the hydrolysate of enzymesoluble pigskin collagen has been successfully separated. The factors affecting the separation are discussed and the optimum separating conditions used in the present study have been selected and established. The result of the separation for the enzyme-soluble pigskin collagen by SDS-page shows that effective separation could be achieved under the experimental conditions of 4% stacking gel and 10% separating gel.
