Since 1959, when cross-linked dextran gels were first prepared by Flodin and Porath, a whole new field of chromatography has become available based on differences in molecular size. The various methods and materials used for gel filtration are described and particular use is made of theoretical considerations to determine the factors which influence resolution during gel filtration. Affinity chromatography on porous media and the use of chemically modified sepharoses are briefly considered.
