The Freeze-Etching Of Skin For Electron Microscope Examination

Abstract

Freeze-etching, a new preparative method for electron microscopy and designed to retain the natural fluid content of the sample, has been investigated for use with fresh skin and with hides at various stages of processing. Cryoprotective agents such as glycerol, which prevent freezing damage, do not appear absolutely necessary for observing the fibrous components of skin. They are however essential for the preservation of the cellular structures. The method offers much scope for studying collagen fibrils in pelt as they are modified under the actual conditions which apply at various stages of leather manufacture
